Tirzepatide 25mg/Pyridoxine 50mg/Glycine 20mg/ml
Comprehensive Scientific Overview
Subject: Enhancing GLP-1 RA Therapy: Mechanistic Rationale for Fixed-Dose Pyridoxine and Glycine
Our new compounded formula ons integrate fixed concentrations of Pyridoxine (B6) and Glycine with your
GLP-1 receptor agonists to address the two major challenges:
Nausea and Muscle Loss during Weight Reduction.
Formula on Breakdown:
• Tirzepatide Formula on: 25 mg/mL Tirzepatide + 50 mg/mL Pyridoxine (B6) + 20 mg/mL Glycine
• Semaglutide Formula on: 2.5 mg/mL Semaglutide + 50 mg/mL Pyridoxine (B6) + 20 mg/mL Glycine
Mechanistic Rationale:
• Pyridoxine(B6) (50 mg/mL):
Nausea Control: Vitamin B6 is a proven antiemetic. Studies in pregnancy have shown that doses as low as
10–25 mg can significantly reduce nausea symptoms. With pyridoxine fixed at 50 mg/mL, even a 0.1 mL
injection delivers ~5 mg B6 per 0.1ml—sufficient to reduce GLP-1–induced nausea as evidenced in clinical trials, while a maximum injection (0.96 mL for semaglutide or 0.6 mL for tirzepatide) delivers 48–30 mg far below toxic levels. Our fixed concentration delivers ~5 mg B6 per 0.1 mL and scales appropriately with dose escalation, ensuring effective nausea mi ga on without risk of overdose.
Metabolic Enzyme Support: B6 is essential in protein metabolism and neurotransmitter production, supporting the body’s energy production during weight loss and helping to preserve lean mass.
Safety: Even at the maximum volume, the delivered B6 remains well below the 100 mg/day upper limit, with toxicity only seen at chronic doses exceeding 200–500 mg/day.
With pyridoxine fixed at 50 mg/mL, even a 0.1 mL injection delivers ~5 mg B6—sufficient to reduce GLP-1 – induced nausea as evidenced in clinical trials, while a maximum injection (0.96 mL for semaglutide or 0.6 mL for tirzepatide) delivers 48–30 mg—far below toxic levels.
Glycine (20 mg/mL):
Muscle Preservation: Glycine has been shown to improve muscle protein synthesis and reduce inflammatory cytokine activity that contributes to muscle catabolism. Research indicates that glycine supplementation improves lean mass retention in weight loss scenarios.
Tolerability and Stability: Glycine fixed at 20 mg/mL provides ~2 mg per 0.1 mL and up to ~19–20 mg at the
highest dose. This ensures an isotonic, supportive dose, ensuring injection comfort, that aids in muscle
preservation without risk, given that therapeutic oral doses are in the gram range. Even at maximum doses,dd the total glycine exposure is minimal relative to safe oral doses used in clinical studies.
